Probiotic Inhibition Effects on Escherichia coli O157:H7 in Yoghurt(ผลของโปรไบโอติกในการยับยั้ง Escherichia Coli O157:H7 ในโยเกิร์ต)

Abstract

การศึกษาผลของโปรไบโอติก 6 ชนิดได้แก่ Lactobacillus acidophilus, Lb. casei, Lb. delbrueckii subsp. lactis, Lb. plantarum, Lactococcus lactis subsp. cremoris และ Lc. lactis IO-1 ในการยับยั้ง Escherichia coli O157:H7 โดยเปรียบเทียบเปอร์เซ็นต์โซนยับยั้งพบว่า Lb. acidophilus และ Lb. plantarum สามารถยับยั้ง E. coli O157:H7 ได้ดีที่สุด จากนั้นนำโปรไบโอติกทั้ง 2 ชนิดมาศึกษาผลการยับยั้ง E. coli O157:H7 ในโยเกิร์ตระหว่างการหมักและการเก็บรักษาโดยใช้จำนวน E. coli O157:H7 เริ่มต้นต่างกัน 3 ระดับคือ 2, 4 และ 6 log cfu/mL อัตราส่วนระหว่างโปรไบโอติกกับเชื้อโยเกิร์ตเท่ากับ 1:1 พบว่า Lb. plantarum มีผลในการยับยั้ง E. coli O157:H7 ได้ดีที่สุดโดยระหว่างการเก็บรักษาสามารถลดจำนวน E. coli O157:H7 ที่มีจำนวนเริ่มต้น 2, 4 และ 6 log cfu/mL จนไม่สามารถตรวจพบได้ภายใน 24, 48 และ 72 ชั่วโมงตามลำดับ และเมื่อนำ Lb. plantarum มาศึกษาผลการยับยั้ง E. coli O157:H7 โดยอัตราส่วนระหว่าง Lb. plantarum และเชื้อโยเกิร์ตแตกต่างกัน 3 ระดับคือ 2:1, 1:1 และ 1:2 พบว่าแต่ละอัตราส่วนมีผลในการยับยั้ง E. coli O157:H7 ไม่แตกต่างกันทางสถิติ (P>0.05) เมื่อนำ Lb. plantarum มาศึกษาผลการยับยั้ง E. coli O157:H7 กรณีมีการปนเปื้อนก่อนและหลังการหมักโดยเปรียบเทียบ D-value ในช่วงการเก็บรักษาพบว่าตัวอย่างที่มีจำนวน E. coli O157:H7 เริ่มต้นเท่ากันเมื่อเติม E. coli O157:H7 ลงในโยเกิร์ตก่อนการหมักจะมี D-value น้อยกว่าตัวอย่างที่เติม E. coli O157:H7 หลังการหมัก โดยสามารถลดจำนวน E. coli O157:H7 ซึ่งเติมลงในตัวอย่างก่อนการหมักจำนวนเริ่มต้น 2, 4 และ 6 log cfu/mL จนไม่สามารถตรวจพบได้ภายใน 24, 48 และ 72 ชั่วโมง และมี D-value เท่ากับ 14.65, 14.77 และ 14.88 ชั่วโมงตามลำดับ ส่วนตัวอย่างที่เติม E. coli O157:H7 หลังการหมักพบว่าสามารถลดจำนวน E. coli O157:H7 ซึ่งมีจำนวนเริ่มต้น 2, 4 และ 6 log cfu/mL จนไม่สามารถตรวจพบได้ภายใน 72, 72 และ 120 ชั่วโมง และมี D-value เท่ากับ 20.35, 18.95 และ 32.79 ชั่วโมงตามลำดับ

Inhibition effects of six strains of probiotics including Lactobacillus acidophilus, Lb. casei, Lb. delbrueckii subsp. lactis, Lb. plantarum, Lactococcus lactis subsp. cremoris, and Lc. lactis IO-1 on Escherichia coli O157:H7 were studied. According to percentage comparison of inhibition zone, Lb. acidophilus and Lb. plantarum were found to inhibit E. coli O157:H7 the most. Then, both types of probiotics were evaluated for inhibition effect on E. coli O157:H7 in yogurt product during fermentation and storage with three different initial concentrations of 2, 4 and 6 log cfu/mL and the ratio of probiotic to yogurt starter as 1:1. It was found that Lb. plantarum can inhibit E. coli O157:H7 the most as it could reduce the E. coli O157:H7 initial count of 2, 4 and 6 log cfu/mL during storage until it could not be detected within 24, 48 and 72 hrs respectively. When studying the inhibition effect of Lb. plantarum on E. coli O157:H7 at the three different Lb. plantarum / yogurt culture ratios of 2:1, 1:1 and 1:2, it was found that there were not significant differences in all ratios (P>0.05); however, in case of pre-and post-fermentation contamination with D-value comparison during storage, it was found that samples with the same initial cell concentration of E. coli O157:H7 had less D-value with pre-fermentation than with post-fermentation as during storage it could reduce E. coli O157:H7 with pre-fermentation artificial contamination at 2, 4 and 6 log cfu/mL until they could not be detected within 24, 48 and 72 hrs with D-values of 14.65, 14.77 and 14.88 hrs respectively while samples with E. coli O157:H7 inoculation at 2, 4 and 6 log cfu/mL after fermentation reduced in numbers of E. coli O157:H7 until they could not be detected within 72, 72 and 120 hrs with D-values of 20.35, 18.95 and 32.79 hrs respectively.
