A Comparative Study on Antioxidant and Antibacterial Activities Against Streptococcus mutans of Five Ocimum Species Extracts Found in Thailand

Abstract
Oral health problems, especially dental caries, are primarily caused by the bacterium Streptococcus mutans (S. mutans), which produces acids and forms biofilms adhering to tooth surfaces, resulting in enamel destruction. Based on this cause, the researchers were interested in studying: 1) the antibacterial activity against S. mutans; 2) the antioxidant activity; and 3) the total phenolic and total flavonoid contents of crude extracts from five Ocimum species, namely Ocimum tenuiflorum, Ocimum sanctum, Ocimum basilicum, Ocimum gratissimum, and Ocimum africanum. The results showed that the crude extract of Ocimum basilicum exhibited the highest antibacterial activity against S. mutans, with a mean inhibition zone of 3.01±0.09 mm. This extract also contained the highest total flavonoid and total phenolic contents at 1.32±0.45 mg QE/g DW and 8.51±0.15 mg GAE/g DW, respectively. Antioxidant activity evaluated by the DPPH radical scavenging assay revealed that the crude extracts of Ocimum sanctum and Ocimum basilicum demonstrated the most prominent antioxidant effects, at 56.32% and 49.19%, respectively. In contrast, the crude extract of Ocimum tenuiflorum showed the lowest total flavonoid and total phenolic contents, as well as the smallest inhibition zone. The percentage yield (% yield) of the crude extracts from the five Ocimum species indicated that Ocimum gratissimum provided the highest percent yield at 12.53±0.74. These findings demonstrate that extracts from certain Ocimum species possess antioxidant and antibacterial activities, which can be further developed into natural products for oral health in the future.
