การเจริญและสร้างฟิล์มชีวภาพของแบคทีเรียสายพันธุ์ Escherichia coli บนพื้นผิวแผ่นสแตนเลสโดยการเพาะเลี้ยงด้วยน้ำนม

บทคัดย่อ

แบคทีเรีย E. coli พบได้ทั่วไปในสิ่งแวดล้อมและส่วนลำไส้ของมนุษย์ ซึ่งเป็นเชื้อฉวยโอกาสและสาเหตุของการติดเชื้อในทางเดินอาหารทำให้เกิดอาการท้องร่วง สามารถรอดชีวิตจากกระบวนการทำความสะอาดด้วยวิธีต่าง ๆ จึงเป็นสาเหตุทำให้มีการปนเปื้อนบนวัสดุและเครื่องมือสำหรับกระบวนการผลิตอาหาร งานวิจัยนี้จึงได้ศึกษาลักษณะการเจริญและการเกิดฟิล์มชีวภาพจากแบคทีเรีย E. coli บนพื้นผิวแผ่นสแตนเลสเกรด 316 แบคทีเรีย E. coli มีลักษณะการเพิ่มปริมาณเซลล์อย่างรวดเร็วทำให้เข้าสู่ช่วงของการเจริญแบบทวีคูณ (Exponential phase) เมื่อเพาะเลี้ยงด้วยอาหารเลี้ยงเชื้อ Nutrient broth (NB) ภายใน 2 ชั่วโมง บ่มที่อุณหภูมิ 37 C (9 log cfu/mL) และมีลักษณะการเจริญทำนองเดียวกันเมื่อเพาะเลี้ยงแบคทีเรีย E. coli ด้วยน้ำนม UHT ในขณะที่การทดสอบเก็บรักษาน้ำนมที่ปนเปื้อนด้วยแบคทีเรีย E. coli ไว้ที่อุณหภูมิ 4 C ยังพบการเพิ่มปริมาณภายใน 2 ชั่วโมง (8 log cfu/mL) เช่นเดียวกัน แต่มีปริมาณแบคทีเรียที่ต่ำกว่า การสร้างฟิล์มชีวภาพเกิดเป็นระยะโครงสร้างสมบูรณ์บนพื้นผิวแผ่นสแตนเลสที่มีแบคทีเรีย E. coli เจริญ ซึ่งเกิดขึ้นได้อย่างรวดเร็วภายใน 2 ชั่วโมง ที่อุณหภูมิ 37 C เมื่อเพาะเลี้ยงด้วยอาหารเลี้ยงเชื้อ NB และน้ำนม UHT แต่การเก็บน้ำนม UHT ที่ปนเปื้อนด้วยแบคทีเรีย E. coli ไว้ที่อุณหภูมิ 4 C มีการเกิดฟิล์มชีวภาพระยะโครงสร้างสมบูรณ์ช้ากว่า (4-6 ชั่วโมง) ดังนั้น การเก็บรักษาน้ำนมด้วยอุณหภูมิต่ำจึงช่วยชะลอการเจริญและการสร้างฟิล์มชีวภาพของแบคทีเรีย E. coli ในช่วงระยะแรก (2 ชั่วโมง) แต่ปริมาณแบคทีเรียไม่ลดปริมาณลงที่อุณหภูมิ 4 C เนื่องจากมีโครงสร้างของฟิล์มชีวภาพช่วยปกป้องเซลล์แบคทีเรียในสภาวะแวดล้อมที่ไม่เหมาะสม
